Description
This method is a quantitative determination of histamine in fish and fish products. The limit of quantitation is 2 mg/kg under the conditions described in this procedure. Histamine is extracted from a homogenized sample with 0.6 M perchloric acid. A specific amount of internal standard is added prior to homogenization. Separation and detection of histamine is performed in a HPLC system with the use of gradient elution, post-column derivatization with o-Phthaldialdehyde (OPA) and fluorescence detection with excitation wavelength at 365 nm and emission wavelength at 418 nm.
