Description
The method has been validated for histamine levels in fish from 5.6 mg/kg to 158 mg/kg. The sample is extracted with 75% (v/v) methanol, and purified on an anion exchange column in order to remove interfering substances. o-Phthaldialdehyde solution is added to the eluate to form fluorescent histamine derivatives. The fluorescent intensity of derivatives is measured spectrophotometrically and histamine is quantified using external standards. This method is endorsed by Codex Alimentarius for the determination of histamine in fish and fish products.
The method has been collaboratively validated by AOAC International and harmonises with AOAC 977.13.
