REPEALED 2022: The Uppsala method provides means to determine dietary fibre content and composition in vegetable containing foods. Starch is removed from the sample using a-amylase and amylo¬glucosidase in acetate buffer. Soluble polymers are precipitated with 80% ethanol. Precipitated and insoluble polysaccharides are hydrolysed with sulphuric acid. Released neutral monosaccharides are quantified by gas chromatography as alditol acetates. Uronic acids in the acid hydrolysate are determined by colorimetry, and Klason lignin is determined gravimetrically as the ash-free acid-insoluble residue. Total dietary fibre is defined as amylase-resistant polysaccharides plus Klason lignin. This is an AOAC-AACC-NMKL method.
